The earliest account of Pedro de Quiroga in the Americas is 1550, at the time when he was a vencino (tax-paying citizen) of Zacatecas in Nueva Galicia. He was listed in a census that was taken in Zacatecas in April 1550. Pedro de Quiroga gave his occupation as herrero (blacksmith) owning some houses.

There is a list that was made late in 1550 that shows the names of vecinos of Zacatecas who established the religious Confradia del Santis mo Sacramento. Pedro de Quiroga appear on this list, which basically is a list of thirty-four men who were among the earliest of settlers of Zacatecas.

Pedro de Quiroga's first wife was named María Ponce de León, and they had two daughters: Luisa Ponce de Le who married Captain Martín Pérez de Uranzua, also known as Pérez de Iranzu; and María Ponce de León who married Antonio Vázquez de Castañeda. Pedro's third daughter, Ana de Gamarda, was an only child by his second, unamed wife, and wife of Juan Borruel de Luna. Records showing the name of hacienda owners of the Valle de la Poana in regard to the distribution of water for the year of 1573 include Juan Borruel de Luna, but Pedro de Quiroga is missing from the list indicating he was possibly dead by that time.
